Nutrient Broth (source: shoormery) Cook peeled and sliced potatoes in mineral water or distilled water on low heat until tender. Filter the broth through a sieve and discard the potatoes.

Potato Broth

  • 500 g potatoes (peeled and sliced thin)
  • 5000 ml water (distilled or mineral water)

Prepare the following nutrient solution using the potato broth prepared above:

Nutrient Solution

  • 200 ml potato broth
  • 2 Gram KH2PO4 (Potassium dihydrogen phosphate)
  • 1 Gram MgSO4 (Magnesium sulphate)
  • 1 g C6H14N2O7 (Ammonium citrate)
  • 30 g glucose (dextrose)
  • 3 g Soybean peptone
  • 50 mg vitamin B1 (thiamine)

Dilute all the above components with water by adjusting the volume to 1000 ml

Measure 35 ml of nutrient solution and pour into wide mouth glass jars. Weigh the rice grains (30g) and put them in the jars along with the nutrient solution. Close the glass flask with a metal lid with a felted disc under the lid. You do not need a hole in the metal lid, only the filter under the lid is enough for the exchange of air during the growth of the mycelium.

Per jar :

  • 35 ml nutrient solution
  • 30 g rice (polished or brown rice)

After sterilization, inject approximately 5 ml of the Liquid culture of Cordyceps militaris into every glass jar (use a micropipette with sterilized tips or even a stainless spoon sterilized on the flame of a Bunsen burner). Incubate in the dark (25ºC) until the mycelium cover all the grains.


Primordia initiation

Start a one week treatment for the initiation of primordia by putting the flasks on the refrigerator at night and leaving them at an illuminated area during the day (18-25 ºC), without direct sunlight. After the appearance of primordia , keep the flasks in a 18-23ºC environment with indirect light until the mushrooms develop to full size.
